Regardless of the mechanism leading to reduced diffusion of the unstimulated phospho-null receptor, this phenomenon, together with the previous observation of some degree of phosphorylation at the WT receptor under vehicle conditions, indicates the existence of a much more dynamic cycling between phosphorylated and unphosphorylated receptor states, present already in the apo-state, which could potentially involve interactions not only with specific GRKs but also with phosphatases that could contribute to the regulation of the overall degree of receptor phosphorylation, explaining why the C-terminal serine phospho-sites have the capacity to influence the behaviour of the receptor even in the absence of agonist stimulation
